REQUIRED INSTRUMENTS AND REAGENTS

  • Microplate reader (450nm wavelength filter).
  • Incubator capable of maintaining 37℃.
  • 1.5mL EP tube, absorbent paper.
  • Pipette and disposable tip: 0.5-10μL, 2-20μL, 20-200μL, 200-1000μL.
  • Double distilled water or deionized water.

ASSAY PROCEDURE SUMMARY

Add 100μL standard or sample to each well, incubate at 37℃ for 90 min. Discard the liquid.

Add 100μL Biotinlated Detection Ab working solution to each well. Incubate at 37℃ for 60 min.
Washing: Aspirate and wash the plate for 3 times.

Add 100μL HRP conjugate working solution. Incubate at 37℃ for 30 min.
Washing: Aspirate and wash the plate for 5 times.

Add 90μL Substrate Reagent. Incubate at 37℃ for 15 min.

Add 50μL Stop Solution.

Readings at 450nm. Calculation of results.

STANDARD CURVE

The product has been verified by the quality inspection department and meets the performance requirements of the product manual. (Laboratory temperature: 20-25℃, laboratory relative humidity: 20-60%. Before the experiment, all components were equilibrated to room temperature.) The following data is for reference only. A standard curve must be run with each test.

RECOVERY

The recovery of Human NSE spiked to three different levels in samples throughout the range of the assay in various matrices was evaluated.

LINEARITY

To assess the linearity of the assay, samples containing and/or spiked with high concentrations of Human NSE in various matrices were diluted with the Reference Standard & Sample Diluent to produce samples with values within the dynamic range of the assay.

PRECISION(%)

Intra-Assay Precision (Precision within an assay): Three samples of known concentration were tested twenty times on one plate to assess intra-assay precision.

Inter-Assay Precision (Precision between assays): Three samples of known concentration were tested in twenty separate assays to assess inter-assay precision. Assays were performed by at least three technicians using two lots of components.
